Viability of Toxoplasma gondii in cattle semen cryopreserved with different concentrations of cryoprotectant

ABSTRACT: Toxoplasma gondii can be eliminated in bovine semen. Cryopreserved semen is often used due to the fact that artificial insemination dairy and beef cattle provides benefits terms of production. However, little known regarding viability infectivity T. tachyzoites cryopreserved In present study, negative for were contaminated with 1 x 106 (RH strain) without different cryoprotectants, such as DMSO (concentrations 2.5%, 5.0%, 7.5%, 8.0% 10.0%) glycerol (2.25%, 3.0%, 7.5% 10.0%), followed by freezing liquid nitrogen (-196°C). After 24 hours, samples thawed inoculated 10 mice per cryoprotectant concentration. The evaluated clinical signs toxoplasmosis (rough coat, diarrhea, hypoactivity sudden death) well serum titers IgM IgG presence peritoneal lavage. results revealed remained infective all samples. Clinical observed beginning 6th day post-inoculation (DPI) 100% lethality was found between 7th 9th DPI.
